Sheriff Jody Mills (Kim Rhodes) arrives at the Minnesota-Dakotas Sheriff's retreat and then tells herself that she can get through the experience. A teenager female beggar approached Jody so the sheriff gave her money and instructed her to buy food to eat for lunch.
At the registration table, Sheriff Mills meets her opposite side, the cheery Sheriff Donna Hanscum (Briana Buckmaster). Another person that Sheriff Mills met was Donna's ex-husband, Doug, who made fun of Donna's weight and then asked for extra meal vouchers.
Donna explains to Jody that she was married to Doug and Jody declares that she gets it. At the Men of Letters bunker, Dean Winchester (Jensen Ackles) tells his brother, Sam (Jared Padalecki), in an annoyed tone that he cannot find anything on the Mark of Cain.
As Sheriff Len Cues welcome the attendees of the retreat, he receives a notification regarding the attack that happened to the male victim in the beginning of the episode in Hibbing, Minnesota. Jody talks to Annie, the girl she helped and decided to take care of in "Supernatural" Season 9, episode 19 titled "Alex Annie Alexis Ann," over the phone.
One task at the retreat is to find a partner and since Sheriff Mills did not find any, Sheriff Hanscum approached her and told her that she now has a partner. While looking at the event schedule, the two find out about the attack that happened and decided to conduct an investigation on it.
Back at the bunker, Sam receives a phone call from Sheriff Mills inquiring about creatures that can eat human bodies. The sheriff told the boys that she can handle the situation but Dean convinces his brother that they should go there, do their own investigation and help Sheriff Mills.
At a bar, Jody and Donna try to talk to Sheriff Cues about the victim but the sheriff insists that animal control is working on the case. Sheriff Cues' deputy arrives to reveal that there is another male victim.
Initially, Sheriff Cues and the deputy tried to keep the information a secret but Sheriff Mills was able to make the sheriff reveal the second case. Donna looks upset that her ex-husband dancing with a younger and sexier sheriff. Sheriff Mills thinks that Donna should just forget him because she thinks he is a "dick."
The two sheriffs tried to bond outside after Donna overheard Jody talking to Alex over the phone. The following morning, Sam and Dean arrive at the retreat to conduct their investigation on the case.
"I said I can handle it," Jody tells the Winchester before hugging them. The sheriff also finds out that Sam and Dean know Sheriff Donna Hanscum after helping her out in "Supernatural" Season 9, episode 13 titled "The Purge." However, Sheriff Hanscum is still oblivious to what Sam and Dean really do.
The Winchesters approached the Hibbing sheriff and his deputy to ask if they can check any surveillance video on the attack. Dean was able to make the deputy reveal that the sheriff has access to the police server and he changed the password right after the first murder.
At the expo, things between Jody and Donna turned sour when Jody finally confronted Donna's ex-husband Doug and told him that he is just a douche. Donna became upset and told Jody that until she lost her husband then she should keep her lips zipped when it comes to her husband.
The comment obviously hit Jody and flashback scenes showed her husband's death. Realizing Donna made a mistake, she apologized to Jody and headed out to get some fresh air.
Outside, Donna noticed a blood trail that led to the third victim, the female sheriff that her ex-husband was dancing with at the bar, and Sheriff Cues was next to her body showing off his vampire fangs. Donna hides in fear that the creature will come after her.
Sam tells Jody and Dean that the footage got deleted and then Donna arrives declaring that she wants to talk to Jody alone. Jody immediately believed what Donna saw and confirmed to the sheriff that monsters do exist out there.
Jody leaves a message on Sam's phone about Sheriff Cues' motel room and the four of them meet up there. Donna is shocked to find out that they are dealing with vampires so Dean told Jody that she should talk to Donna about the truth.
Sam, Dean, Jody and Donna headed to the address that Donna uncovered at Sheriff Cues' motel room. Sam sees the Sheriff Cues inside an old farm barn. The sheriff warned them that they should run but it is already too late as a group of vampires, led by the female beggar that Jody gave lunch money, knocked out and tied up the female sheriffs and the Winchesters.
It is revealed that Len Cues was part of the girl's vampire nest but he walked away, started a new life as a sheriff and fed through blood bags only. Sheriff Cues explained that what Donna saw was himself trying to fight the urge from drinking blood or attacking someone after smelling the victim's blood.
Sheriff Cues agreed to go back to the nest if it means the vampires will not kill Sam, Dean, Jody and Donna but the girl instructed the sheriff to kill them instead. When the sheriff refused, the girl chopped of his head.
Dean and Donna were able to break free from being tied up. Dean fights off the other vampires while Donna decapitated the girl leader when the teenager vampire scavenger tried to kill Sheriff Mills. "Hakuna Matata, lady," Sheriff Donna Hanscum declared after experiencing her first hunt and kill.
The ending scene showed a Winchester brotherly talk where Dean tells Sam that he did not feel the effects of the Mark of Cain while killing the vampires but Sam knows that his brother is lying. As for Donna, the sheriff admitted to Jody that she now has open eyes to the darker and more monstrous world. Sheriff Mills offered her help, particularly with her supernatural knowledge, to the newbie in the monster world.
